Elements in support of the 'non-identity' of the PGRMC1 protein with the σ<inf>2</inf> receptor

Abstract

σ2 Receptorsubtypeisoverexpressedinavarietyofhumantumors,with σ2 agonists showing antiproliferativeeffectstowardstumorcellsthroughmultiplepathwaysthatdependbothonthetumor cell typeandonthemoleculetype.Therefore, σ2 receptorisanintriguingtargetfortumordiagnosisand treatment despitethefactthatthatithasnotyetbeencloned.Oneofthelastattemptstocharacterize σ2 receptorsledtoidentifyitastheprogesteronereceptormembranecomponent1(PGRMC1).Although still controversial,suchidentityappearstohavebeenaccepted.Wetheaimofcontributingtosolvethis controversy,inthisworkwestablysilencedoroverexpressedPGRMC1proteininhumanMCF7 adenocarcinoma cells.WesternblottinganalyseswereperformedtoquantifythepresenceofPGRMC1 proteinoneachofthethreeMCF7celllinesvariants,whilescatchardanalyseswithradioligandwere performed inordertodeterminetheexpressionofthe σ2 receptors.Inordertocorrelatethe antiproliferativeeffectof σ2 receptoragonistwithPGRMC1density,some σ2 ligands wereadministered to eachofthethreeMCF7cellsvariants.TheresultssuggestedthatPGRMC1and σ2 receptorsaretwo different molecularentities.
