Quartz crystal microbalance with dissipation (QCM-D) as tool to exploit antigen–antibody interactions in pancreatic ductal adenocarcinomadetection

Abstract

Novel syntheticpeptidesrepresentsmartmoleculesforantigen–antibodyinteractionsinseveral bioanalyticsapplications,frompurificationtoserumscreening.Theirimmobilizationontoasolid phase isconsideredakeypointforsensitivityincreasing.Inthisview,weexploitedQuartzCrystal Microbalancewithsimultaneousfrequencyanddissipationmonitoring(QCM-D)withadoubleaim, specifically,asinvestigativetoolforspacersmonolayerassemblinganditsfunctionalevaluation,as well ashighsensitivemethodforspecificimmunosorbentassays.Themethodwasappliedto pancreaticductaladenocarcinoma(PDAC)detectionbystudyingtheinteractionsbetweensynthetic phosphorylatedandun-phosphorylated a-enolasepeptideswithseraofhealthyandPDACpatients.The syntheticpeptideswereimmobilizedonthegoldsurfaceoftheQCM-Dsensorviaaself-assembled alkanethiolmonolayer.Thepresentedexperimentalresultscanbeappliedtothedevelopmentof surfaceslesssensitivetonon-specificinteractionswiththefinaltargettosuggestspecificprotocolsfor detectingPDACmarkerswithun-labeledbiosensors.
